    Enhancing performance in anaerobic high-solids stratified bed digesters by straw bed implementation

    Anaerobic high-solids single-stage stratified bed digesters have been found to be simple and flexible design candidates for small-scale reactors located in medium- to low-technology environments. In the present study, wheat straw was used as the starter material for the stratified bed. Upon green mass feeding, the anaerobically stabilised straw bed functioned both as a biofilm support and as a particulate filter. It enabled a direct onset of 7 kg VSm(-3) batch loads, added twice a week, and permitted a low but consistent bed permeability during feeding at an average superficial flow velocity of 1 m d(-1) to be achieved. Fed-batch tests with sugar beet tops in pilot- and laboratory-scale setups at an average loading rate of 2 kg VSm(-3) d(-1) resulted in average biogas production rates of 1.2-1.4 m3 m(-3) d(-1) and methane yields of 0.31-0.36 m3 kg(-1) VS(added). At the end of the laboratory-scale feeding trial, the 200 day old straw bed had compacted to 50% of its initial volume, without any negative effects on performance being detectable

    Sheet-metal press line parameter tuning using a combined DIRECT and Nelder-Mead algorithm

    It is a great challenge to obtain an efficient algorithm for global optimisation of nonlinear, nonconvex and high dimensional objective functions. This paper shows how the combination of DIRECT and Nelder-Mead algorithms can improve the efficiency in the parameter tuning of a sheet-metal press line. A combined optimisation algorithm is proposed that determines and utilises all local optimal points from DIRECT algorithm as Nelder-Mead starting points. To reduce the total optimisation time, all Nelder-Mead optimisations can be executed in parallel. Additionally, a Collision Inspection Method is implemented in the simulation model to reduce the evaluation time. Altogether, this results in an industrially useful parameter tuning method. Improvements of an increased production rate of 7% and 40% smoother robot motions have been achieved

    Sulfide in engineered methanogenic systems - Friend or foe?

    Sulfide ions are regarded to be toxic to microorganisms in engineered methanogenic systems (EMS), where organic substances are anaerobically converted to products such as methane, hydrogen, alcohols, and carboxylic acids. A vast body of research has addressed solutions to mitigate process disturbances associated with high sulfide levels, yet the established paradigm has drawn the attention away from the multifaceted sulfide interactions with minerals, organics, microbial interfaces and their implications for performance of EMS. This brief review brings forward sulfide-derived pathways other than toxicity and with potential significance for anaerobic organic matter degradation. Available evidence on sulfide reactions with organic matter, interventions with key microbial metabolisms, and interspecies electron transfer are critically synthesized as a guidance for comprehending the sulfide effects on EMS apart from the microbial toxicity. The outcomes identify existing knowledge gaps and specify future research needs as a step forward towards realizing the potential of sulfide-derived mechanisms in diversifying and optimizing EMS applications

    Health-related quality of life, utility, and productivity outcomes instruments: ease of completion by subjects with COPD

    An important outcome of any clinical intervention is the change in the subject's own perceived state of health. This can be categorized as health-related quality of life (HRQL), utility (preference-based health state), and daily life performance. 174 Swedish subjects with chronic obstructive pulmonary disease (COPD) (mean age 64.3 ± 12 years) completed five self-administered questionnaires: Short Form 36 (SF-36), St George's Respiratory Questionnaire (SGRQ), EuroQol-5D (EQ-5D), Health States-COPD (HS-COPD), and Work Productivity and Activity Impairment Questionnaire for COPD (WPAI-COPD). The subjects scored these outcomes instruments for ease of completion using a 5-point scale. The time taken to complete them was noted and the administrators' opinion of the subjects' comprehension of the questionnaires recorded using a 4-point scale. A score of 1–3 ("very easy" to "acceptable") was recorded by 92% of subjects for the SF-36, 90% for SGRQ, 80% for EQ-5D, 83% for WPAI-COPD, and 53% for HS-COPD. The HS-COPD was graded "very difficult" to complete by 21% of subjects compared with 3–5% of subjects for the other questionnaires. The mean time taken to complete all questionnaires was 39 minutes, and the large majority of subjects scored "good" for understanding by the administrator. Age correlated significantly with the degree of the subject's opinion of the ease of completion of five outcomes instruments, while the influence of gender, socio-economic status and disease severity was not statistically significant

    Effect of Cobalt, Nickel, and Selenium/Tungsten Deficiency on Mesophilic Anaerobic Digestion of Chemically Defined Soluble Organic Compounds

    Trace elements (TEs) are vital for anaerobic digestion (AD), due to their role as cofactors in many key enzymes. The aim of this study was to evaluate the effects of specific TE deficiencies on mixed microbial communities during AD of soluble polymer-free substrates, thus focusing on AD after hydrolysis. Three mesophilic (37 degrees C) continuous stirred-tank biogas reactors were depleted either of Co, Ni, or a combination of Se and W, respectively, by discontinuing their supplementation. Ni and Se/W depletion led to changes in methane kinetics, linked to progressive volatile fatty acid (VFA) accumulation, eventually resulting in process failure. No significant changes occurred in the Co-depleted reactor, indicating that the amount of Co present in the substrate in absence of supplementation was sufficient to maintain process stability. Archaeal communities remained fairly stable independent of TE concentrations, while bacterial communities gradually changed with VFA accumulation in Ni- and Se-/W-depleted reactors. Despite this, the communities remained relatively similar between these two reactors, suggesting that the major shifts in composition likely occurred due to the accumulating VFAs. Overall, the results indicate that Ni and Se/W depletion primarily lead to slower metabolic activities of methanogenic archaea and their syntrophic partners, which then has a ripple effect throughout the microbial community due to a gradual accumulation of intermediate fermentation products
